This sounds a little silly and you probably know already but sometimes I go the "playfinder" function at dramatists.com, type in a desired cast size and just browse through what's available.

 Or Doollee.com
Or Playscripts .com and I think most script sites have that function.

I invite you to read the first act of my comedy, "One Before Forty," online at http://johnshanahan.net  3 men, 2 women, runs about 75 minutes. Minimal set. It's been produced twice now to appreciative audiences! If you like what you see, email me for a complete script.

I've done Samuel French & Dramatists, but nothing really jumps out.

Barefoot in the Park has a cast of five (plus one non-speaking extra) and holds up pretty well.

Try these for three men and two women or these for three women and two men (Because of doubling, there is some overlap between the two lists.) You don't say how long a show you are looking for, so that's a mixture of one-acts and full length plays. Amongst the first list, I would pick out "Merely Players" because it does something very theatrical. (It's an English script, which may be a drawback for you - therefore some of the others might be a better fit.)

How about a new comedy by Philip Ayckbourn. If that last name is familiar, it's because he's Sir Alan Ayckbourn's son! The Essence of Love (3m, 2f), full length comedy A mother and her daughter together with a father and his son find themselves on adjacent hotel terraces in Marrakech. There’s anything but love in the air for this four on vacation … and when the gift of a magical elixir gets into the water, unlikely passions take root. This promises to be no ordinary spell in the sun! Overbearing Diana with her need for drama; her anaemic, idealist daughter, Gemma; the aloof, young artist, Tom and his embarrassing, eternal skirt-chasing father, Martin, are in for a vacation they won’t forget! ‘The Essence of Love’ looks at the longings of the heart and what remains when passion has run its course.
